The selection of highly specific and sensitive antibody is the major part of developing antibody-based detection assays for diagnosing viral pathogens. The western blot is considered the gold standard for analyzing antigen-antibody reaction. Here, the most common and highly used western blot protocol is visualized. After virus isolation, the protein of interest is obtained with a lysis passage. Then, western blotting is performed and the isolated protein is visualized through antibody probing directly on the PVDF membrane.
