Description
This illustration shows that after PAM binding, the REC1 domain on the REC lobe (recognition lobe) moves closer to the PI domain on the Nucleating lobe. This is done by the Bridge Helix (BH) and causes the Target DNA to overlap bend 45 degrees up from the PAM site (which is bound to the PI domain on CRISPR-Cas9). The DNA now overlaps the single guide RNA and this then causes R-loop formation to occur. If R-loop formation is successful then the nucleating sites are activated and cleave the tDNA
