Poly(ADP-ribosyl)ation is a post-translational modification synthesized by poly(ADP-ribose)polymerases (PARP) and catabolized mainly by poly(ADP-ribose)glycohydrolase (PARG). Unlike higher eukaryotes, T. cruzi has only one PARP and one PARG. When DNA damage is detected, TcPARP translocates to the nucleus and transfer ADP-ribose moieties from NAD+ to acceptor proteins and PARP itself. PAR is dynamically removed by PARG. When massive genotoxic damage is detected, PAR can act as a death signal.
