Plaque-forming assay
Description
Visualization of the plaque forming assay. To calculate the viral titre, ten-fold dilutions of an unknown quantity of virus are prepared and then inoculated into susceptible cells (monolayer) and the monolayer is covered with an immobilizing overlay. After incubation, the monolayers are stained and the plaques counted. Only plates with 10 to 100 plaques are counted. The viral titre (PFU/mL) is calculated via the formula.
