Description
Paclitaxel, derived from Taxus brevifolia, is a tetracyclic diterpenoid. It targets the taxane site located within the interior of tubulin, where microtubules face the lumen. Its mechanism of action on microtubules is unique to the compound. Paclitaxel and similar drugs stabilize tumor-associated microtubules through an allosteric mechanism, inhibiting dimer compaction. There is a significant response to chemotherapy, particularly noticeable in the β-III and β-I microtubule populations.
