Metastasis to Bone Disrupts Bone Homeostasis

Description

This template was adapted from the original submission. Edits were made to enhance scientific accuracy, optimal usability and/or to meet industry-leading design standards for science communication. Bone homeostasis balances bone building by osteoblasts (OBs) and absorption by osteoclasts (OCs). Bone is a common metastatic site for breast cancers (BCs). BC cells induce OBs to secrete RANKL, which bind RANK on OCs increasing OC proliferation. Increased OC activity elicits greater resorption, releasing pro-tumorigenic growth factors. This vicious cycle causes bone loss and BC growth. Drugs break this cycle by inducing OC apoptosis (bisphosphonates) or inhibiting OC proliferation (denosumab).
