Enrichment process of S-sulfhydrated proteins from cell lysates using biotin-HPDP
Description
This template depicts the method to enrich S-sulfhydrated proteins from cell lysates. The cells are lysed by passing through a 26-3/4-gauge needle multiple times. The thiol groups of free cysteines are blocked using MMTS. Unused MMTS is removed by protein precipitation using acetone. The proteins are resuspended and labelled using biotin-HPDP. The biotin-labelled proteins are pulled down using streptavidin agarose beads, after which the proteins are extracted by denaturation.
