Direct and Indirect Coombs Tests (DAT and IAT)
Description
To determine the presence of RBC-targeting antibodies, a Coombs test is used. Direct Coombs tests (DAT) use RBCs collected from a patient, typically a neonate. The RBCs are mixed with anti-human IgG. If the sample agglutinates, the RBC-targeting antibodies are present. In the Indirect Coombs Test (IAT), reagent RBCs, usually Rh+, are mixed with the patient's serum before mixing with anti-human IgG. If the sample agglutinates, the RBC-targeting antibodies are present.
