Calcein-AM Efflux Assay
Description
Calcein-AM, a non-fluorescent substrate of ABCB1, is enzymatically converted into green calcein inside cells if not effluxed. However, if cells do express ABCB1, this protein promotes the efflux of calcein-AM, reducing green calcein production. Efflux inhibitors like verapamil can retain calcein, resulting in green fluorescence, indicating functional ABCB1 activity in cells.
