ATAC sequencing is a technique that utilizes Tn5 transposase to insert adapters into open chromatin regions within the genomes of live cells. These adapters are then amplified by PCR and sequenced, to identify the open chromatin sequences and their locations in the genome. This technique allows for the analysis of chromatin accessibility at a genome-wide scale.
